Labelling of human and Schistosoma mansoni hypoxanthine-guanine phosphoribosyltransferases (HGPRTases) with GMP-2',3'-dialdehyde (ox-GMO) resulted in nearly complete inactivation of the enzymes. Digestion of the x-GMP-modified HGPRTases with trypsin followed by HPLC fractionation, partial amino acid sequencing and mass spectral analysis of the labelled peptides revealed that 4 peptides from each HGPRTase were labelled with ox-GMP.
