Vegetative buds from young rhizome of Alpinia calcarata, a medicinal perennial, were cultured on different basal media, such as MS, B-5 and White's medium, for direct shoot induction. Maximum number of shoots (6-9) were obtained from rhizome bud explant on MS medium supplemented with Benzylaminopurine (1.5 mg/l) and Indole-3-acetic acid (0.5 mg/l). Effect of different cytokinins, singly and in combination with auxins, on shoot multiplication were also studied. The shoots were healthy with dark green leaves and attained an average length of 7-10 cm within 50 days. The growth and multiplication rate were directly related with the number of subculture. These multiple shoots produced roots from the basal region in the same media after 20 days. About 80% of the plantlets survived under field conditions.
