A gene TCDTAN that can express a minus-strand diphtheria toxin A chain (DTA) RNA with 631 nucleotides upstream of the tobacco mosaic tobamovirus coat protein start codon, in front of DTA open reading frame (ORF) in cells, was constructed and co-transformed with chloramphenicol transferase (CAT) gene DNA and TMV-RNA into tobacco protoplasts, resulting in inhibition of CAT gene expression at protein level.
