Plasmids, derivatives of ColE1, fail to replicate in bacteria not belonging to the genus Escherichia. Chromosomal mutations of Agrobacterium radiobacter 5D-1, which "afford" replication of ColE1 plasmid in cells of this soil saprophytic bacterium belonging to the Rhizobiaceae family, were identified. These mutations were obtained as a result of plasmid pSUP2021 integration into the chromosome of Agrobacterium.
