Cryopreservation of apple in vitro cultured shoot tips

Shoots tips were excised from 70-day-old in vitro cultured plantlets of Malus domestica cultivars Tsugaru, Orin, Red Fuji, Jonagold, HAC9 and Senshu and M. robusta, and then cold-hardened at 5 degrees C for 3 weeks. After preculture in 0.1-0.7 M sucrose, shoot tips were encapsulated with 3% Na-alginate and 100 mM CaCl2. The beads of 4-5 mm diameter were dehydrated in airflow for 4 h and preserved in liquid nitrogen.
