Somaclonal variation in plants regenerated from callus and protoplasts of taro cv. 'Yatsugashira'

Many green compact cell masses (calloids) and a small number of yellow friable calluses were formed by culturing segments of etiolated stem of Colocasia esculenta Yatsugashira on MS medium supplemented with 30 g sucrose, 1 mg 2,4-D and 1 mg 2iP (isopentenyladenine)/litre. Friable calluses were proliferated by subculturing on the same fresh medium, and used to produce suspension cultures in a liquid medium. Protoplasts were isolated from these suspension cells.
