Abstract
A 1134 bp defective replicase gene of the GPV isolate of barley yellow dwarf virus was amplified by RT-PCR. The amplified product was cloned into pUC19 at the SmaI site and sequenced. The defective replicase gene was inserted into the plant expression vector pPPI8. Common wheat varieties Longjian 127, Shan 160 and Jinmai 47 were transformed by pollen tube pathway. The results of kanamycin-resistant selection, PCR and PCR-Southern test showed that transgenic plants for all the three varieties were obtained.
