The new plasmid pXT3sacB was constructed from pJBJ106, pBluescript SK(+) and pUCD800. The wild-type T-DNA of Agrobacterium rhizogenes K599, which induces hair roots, was deleted using pXT3sac by homologous recombination. A disarming K599 contains selective markers for resistance to ampicillin/carbenicillin, and the growth inhibition by 10% sucrose.
