The objective of this study was to optimize the HPLC-FD (fluorescence detection) method used for detecting okadaic acid in shellfish. The substitution of N,N-diisopropylethylamine by pyridine, ammonium hydroxide or triethylamine for the derivatization of okadaic acid with 1-(bromoacetyl) pyrene was tested. These possible substitutes were used as either pure or diluted with acetonitrile at different concentrations, with different reaction times at 75+or-1 degrees C.
