A specific and sensitive DNA probe was prepared for use in the detection of Pneumocystis carinii DNA in the lungs of infected rats and in clinical specimens from patients with P. carinii pneumonia. The large subunit mitochondrial ribosomal RNA gene of P. carinii was amplified by polymerase chain reaction (PCR) and used as a template. Digoxigenin labelled non-radioactive probe was prepared by random priming.
