1 2 di methyl hydrazine induced colonic tumors in rats

1,2-Dimethylhydrazine [1,2-DMH] was injected s.c. on a weekly basis of 15 mg/kg, for periods of 1-28 wk, into 500 Wistar rats. Glandular mucin depletion appeared as early as the 2nd injection (sulfomucins disappeared). Dysplasia appeared from the 6th wk; if the animals were allowed to survive for a longer period they developed colonic carcinomas, mainly in the left colon. This dysplasia, which was first mild, then moderate, finally became severe in rat mucosa or in the area of polyps. Dysplastic polyps became less frequent when carcinomas increased in number and in size. The average length of survival after cessation of the exposure was inversely proportional to the total dose administered. The glandular mucin depletion-dysplasia-carcinoma sequence characterizes the types of formation of 1,2-DMH-induced colonic carcinomas. One of the striking features was the early appearance of this sequence.
