A rapid high performance liquid chromatographic method for separating tocopherols

A high-performance liquid chromatographic method that uses a short column (100 .times. 4.6 mm) packed with 3-.mu.m silica particles for separation of tocopherols is reported. Baseline separation of .alpha.-, .beta.-, .gamma.-, and .delta.-tocopherols was obtained under isocratic conditions using 0.2% (v/v) 2-propanol in hexane as the mobile phase at a flow rate of 2 ml min-1. Chromatographic separation of tocopherols on this short silica column was compared to that obtained on conventional columns (column length 250 or 300 mm) packed with either pellicular material or 5- or 10-.mu.m silica particles. The short column with 3-.mu.m packing was equally effective in separating tocopherols but substantially reduced analysis time and quantity of solvent used compared with conventional columns.
