Acid phosphatase alkaline phosphatase and glucose 6 phosphate dehydrogenase activities in the kidney and adrenal gland of gamma irradiated rats

The activities of acid phosphatase, alkaline phosphatase and glucose-6-phosphate dehydrogenase (G-6-PD) were studied cytochemically in the kidney and adrenal gland of normal and gamma-irradiated albino rats. In the 2 main zones of the control rat kidney (cortex and medulla), acid phosphatase activity was highest in the cortex than the medulla due to the strong positive reaction exhibited by the lysosomal enzyme present in the proximal convoluted tubules. This order was the same as that found for alkaline phosphatase and G-6-PD. Following .gamma. irradiation, there was a slight increase in the acid phosphatase activity in the proximal convoluted tubules in the cortex. A significant increase in alkaline phosphatase activity was noticed in the cortex, especially in the proximal convoluted tubules and the glomeruli. The activity of G-6-PD was greatly decreased in many of the proximal convoluted tubules in the cortex. In the adrenal cortex, the acid phosphatase-containing particles, lysosomes, were abundant in the zona reticularis and the inner region of the zona fasciculata and also in the cells of the adrenal medulla of control rats. In .gamma.-irradiated rats, lysosomal particles were increased in the 3 zones of the adrenal cortex: zona glomerulosa, zona fasciculata and zona reticularis and in the adrenal medulla. Alkaline phosphatase was elevated in the 3 zones of the adrenal cortex and was depressed in the adrenal medulla. The glycolytic enzyme G-6-PD was inhibited in the adrenal cortex. This enzyme was very low in the adrenal medulla of both normal and .gamma.-irradiated rats.
