A radioimmunoassay (RIA) for the amino terminal region of the bovine parathyroid hormone (B-PTH) fragment has been developed by iodination of synthetic 1-34 bovine parathyroid hormone molecule and using amino terminal recoginizing B-PTH antiserum which was made in a guinea pig. The carboxyl terminal of B-PTH RIA was carried out by iodinating native 1-84 B-PTH and a B-PTH antiserum, which mainly recognize the carboxyl terminal of B-PTH, a reported previously by the authors' laboratory. The data obtained from both amino and carboxy terminal B-PTH RIA were compared among three groups of cows, (1) parturient hypocalcemic, (2) nonparetic parturient, and (3) ethylenediaminetetraacetic acid (EDTA) infused cows. It was observed that plasma levels of amino terminal of B-PTH indicating biologically active B-PTH were only 13.9% of the carboxyl terminal of B-PTH in group 1. There was a significant difference between groups 1 and 2 in carboxy terminal RIA. In the results of the amino terminal B-PTH RIA system, however, no significant difference was observed. On the other hand, the amino terminal of B-PTH molecules was 74.9% of the carboxyl terminal of B-PTH molecules in experimentally induced (EDTA infused) hypocalcemic cattle. The results obtained from this experiment suggest that amino terminal of the B-PTH molecule RIA system may give better evaluation on differential secretion change of parathyroid function at the time than carboxyl terminal of the B-PTH RIA system especially under conditions severe hypocalcemic conditions, such as those observed in parturient paretic cows.
