Basidiospore germination of tricholoma matsutake ii. evaluations of germination conditions and microscopic observations of germination stages

Effects of several fundamental conditions on basidiospore germination of Tricholoma matsutake were examined and the germination stages were observed by microscopy. Half of the basidiospores maintained the ability to germinate for about 35 days when stored at -19.degree. C in the Petri dishes containing with water. The optimum pH for basidiospore germination was 4.3 to 4.9 and the temperature was 20.degree. to 23.degree. C, on agar media containing 0.05 ml/l butyric acid. Swelling of the basidiospores started within a day of inoculation, after 15 days, the diameters of the basidiospores were 8.1 .mu.m, 1.3 times the size before inoculation. The germination rate began to increase on the 2nd day and reached a maximum 10 to 15 days after inoculation. One germ tube generally emerged from one spore. The diameters of germ tubes in the early stage of germination were greater than those of cultured hyphae isolated from the fruit-bodies and they became smaller as incubation progressed.
