Microspectrophotometric measurements of the absorption spectra of guard cells of C. communis and A. cepa indicate a high absorbance in the blue region of the spectrum with a maximum at 340 nm. Maximum blue light absorption occurred in the polar regions of the guard cells, with significantly lower absorption in the subsidiary and epidermal cells. Measurements of absorbance after plasmolysing and deplasmolysing the guard cells indicated that maximum blue light absorption occurred within the cytoplasmic or vacuolar contents of the guard cells. Preliminary chromatographic analysis of epidermal pigments indicated the presence of considerable quantities of flavonoids which absorb light in the UV and blue region of the spectrum. These data are discussed in relation to the enhancement of stomatal opening by blue light.
