Characterization of the cellulase and hemicellulase complexes of geotrichum candidum 3c and of the mutant strain geotrichum candidum 3c 106

A comparative study was performed of the activities and the composition of the enzymes splitting cellulose and hemicellulose (xylans, galactans, galactomannans, arabinans), as well as of glucosidase found in the culture filtrate of the fungus Geotrichum candidum 3C and its mutant strain 3C-106 got by induced selection. No correlation was revealed between cellulose and hemicellulase activities of the strains under study. The difference in the amount and acid-base properties of multiforms of the enzymes from the initial and mutant strain was found by isoelectric focusing. G.candidum 3C-106 produced at least four endo-1,4-.beta.-D-glucanases, three cellobiases and an enzyme that together with endoglucanase and cellobiase decomposed to sugar the cotton fibre cellulose, two galactanases, two mannanases, five endo-1,4-.beta.-D-xylanases, three arabinases, four .beta.-D-xylosidases, two .alpha.-D-galactosidases, two .beta.-D-galactosidases, and four .beta.-D-glucosidases. G. candidum 3C was found to produce at least two endoglucanases, two cellobiases, and an enzyme splitting the cotton fiber cellulose into sugar, one galactanase, one mannase, two arabinases, one endoxylanase, three .beta.-D-xylosidases, one .alpha.-D-galactosidase, two .beta.-D-galactosidases and two .beta.-D-glucosidases.
