Comparative study of the molecular weight of the structural proteins from rabies strains using poly acrylamide gel electrophoresis

A comparative study of the structure of proteins from 5 rabies virus strains (CVS, Flury-HEP, ERA, Pasteur and Dikovania) and Obodhiang virus was carried out using polyacrylamide gel electrophoresis in the presence of SDS (sodium dodecyl sulfate) and 2-mercaptoethanol after concentration and purification of the viruses. The MW of the M1 envelope protein and glycoproteins differed between the 5 strains. There is little difference between the other structural proteins (N, M2 and L). All strains had some minor proteins between L and G and between N and M1. Two of the minor proteins between L and G are probably located on the virion surface because they disappear, as do glycoproteins, after treatment of the virus with trypsin. One of these minor proteins may be a glycoprotein dimer. There is no difference between the 4 major proteins of complete and defective virus. There is little difference in MW betwen the 4 major polypeptides of the Pasteur strain when cultivated on 4 different cell types.
