Comparison of cytotoxicity mediated by human monocyte-macrophages and lymphocytes after prolonged in vitro incubation

The effect on cytotoxicity of in vitro incubation of lymphocytes (L) and monocyte-macrophages (MP) for 2 or 24 h, together (unseparated) or separated, was examined. The yield of MP decreased with prolonged incubation while similar numbers of viable L were obtained with 2 and 24 h incubation. L and MP spontaneous cytotoxicity (SCT) declined with prolonged incubation. L ADCC [antibody-dependent cellular cytotoxicity] increased after 24 h incubation of separated and unseparated cells. MP ADCC increased only after incubation of L and MP together and not when MP were incubated separately for 24 h. Circulating MP ADCC effector cells and possibly L ADCC effector cells may be functionally immature, and by interaction with non-adherent mononuclear cells can be made more active or more numerous.
