Control of flux by isozymes and alternate enzymes application to horse liver alcohol dehydrogenase

The equations required to determine the enzyme/isozyme flux-control coefficients for reactions catalyzed by three or more isozymes or different enzymes (parallel pathways) are derived. The equations are then used to calculate the flux-control coefficients for the three major isozymes of horse liver alcohol dehydrogenase (E.C. 1.1.1.1), EE, ES and SS, and aldehyde dehydrogenase (E.C. 1.2.1.3). If all of the acetaldehyde is metabolized by the F1 cytosolic isozyme of aldehyde dehydrogenase under the assumed cellular conditions then 87% of the control of flux is exerted by aldehyde dehydrogenase and 9% by the EE isozyme of alcohol dehydrogenase. If all of the acetaldehyde is metabolized by the F2 mitochondrial isozyme of aldehyde dehydrogenase 54% of control of flux is exerted by aldehyde dehydrogenase and 32% by the EE isozyme of alcohol dehydrogenase.
