Culture of in vitro fertilized bovine embryos in a chemically defined medium bo medium

Bovine oocytes matured in vitro in TCM-199 were inseminated with semen which had been precultured for 5 h and 6 h after insemination they were cultured in a chemically defined medium, BO-medium (Brackett and Oliphant, 1975) supplemented with 10% IC-SS (immediately centrifuged steer serum) or Ham;s F10 medium supplemented with 10% IC-Ss for further development. The proportion of oocytes developed to the 8-cell stage 60 h after insemination was 4.2% (6/142) and 0.8% (1/26) when they were cultured in Ham's F10 medium and in BO-medium, respectively. The development of oocytes to the 8-cell stage was improved up to 5% in rate when the cumulus cells were removed 30 h after insemination during the process of 60 h culture in BO-medium. The cumulus cells were removed 30 h after insemination and the rate of cleaved embryos were examined. The rates of embryos developed up to the 2-cell stage were 17.6% (55/312) in Ham's F10 medium and 14.9% (46/308) in BO-medium, 30 h after insemination. The rates of 4-cell stage embryos were 2.9% (9/312) in Ham's F10 and 1.3% (4/308) in BO-medium 36 h after insemination. The rates of 8-cell stage embryos were 3.5% (11/312) in Ham's F10 and 4.2% (13/308) in BO-medium. In vitro fertilized oocytes that had been cultured in BO-medium for 60 h and developed to the 5- .apprx. 8-cell stage embryos were transferred into rabbit oviduct. The embryos recovered 100 .apprx. 108 h after transfer had developed to the blastocyst stage.
