Cyto photometric study of dna content in the nuclei of spermatogenic cells of di ploid and poly ploid silkworms

The intensity of Feulgen-DNA staining in the nuclei of spermatogenic cells of diploid and polyploid silkworms [Bombyx mori, B. allotetraploideus] was measured cytophotometrically. Relations of the amount of Feulgen-DNA staining in the nuclei of spermatogonia, primary spermatocytes, spermatids and spermatozoa of diploids were near the theoretical ratio 2:4:1:1. Among spermatogenic cells in diploids, single atyptical cells were observed with a lesser Feulgen-DNA content than in the typical ones. These were termed oligopyrene spermatogenic cells. Tri-and allotetraploid silkworms showed greater variability in the amount of Feulgen-DNA staining in all classes of spermatogenic cells as compared to diploids. Their cell populations were heterogeneous and did not correspond to the normal distribution due to the availability of a great number of oligopyrene and hyperpyrene spermatogenic cells. The results of the quantitative measurements agreed with recent data on the ultrastructural study of silkworm spermatogenesis. The atypical character of development may originate from the stages of spermatogonial divisions.
