The cytophotometric content of DNA in chick embryo hepatocytes was studied and compared to nuclear sizes in various phases of incubation. A 1.5-1.7 increase of nuclear size was accompanied by a slight (10%) increase of DNA content. A significant increase of nuclear size in the liver of chick embryo appears to occur at the onset of the S phase of the mitotic cycle. This facilitates the identification, according to size, of nuclei at the onset of the S phase from curves of DNA content dependence on nuclear area. On the histogram of DNA distribution this part of the nucleus merges with the peak of diploid nuclei.
