Cytochemical investigation of acetyl serotonin transferase activity in the pineal gland

A new method was established for a patent EM investigation of acetyl serotonin transferase. The copper-ferricyanide method revealed a mitochondrial localization in the rat pineal gland. The powerful electron-scattering reaction byproduct was situated on the outer and inner membrane as well as in the inner compartment of the mitochondrium. HgCl2 completely blocked the enzyme. Without serotonin or acetyl-CoA the reaction was negative. No extramitochondrial artefact-reaction was observed. The light-dark cycle biological rhythm apparently has no distinct effect on the cytochemical reaction acivity.
