Detection of hepatitis b surface antigen in liver tissue by a modified immunofluorescent technique and the peroxidase antiperoxidase method

This paper discusses the possibilities of applying a modified immunofluorescent technique (IIF) and the PAP (peroxidase-antiperoxidase) method for the detection of HBsAg in conventional formalin-fixed, paraffin-embedded liver tissue. A specific complex of antibody-HBsAg/HBsAb- HBsAg/ clearly visible in the cytoplasm of hepatocytes were obtained after employing both techniques. The detection of HBsAg in liver tissue by the modified IIF technique was improved by trypsinization. A multilayering immunoenzyme PAP method made it possible to demonstrate viral antigens and distinguish histological details. By properly preparing the tissue and using immunobiological reagents under optimal conditions, complete elimination of nonspecific staining was achieved and the obtained results were reliable and reproducible.
