Development of a serological technique for the early diagnosis of hepatic lesion

Hepatic-specific antigens (HSA) have been isolated, purified and detected in the sera of humans and animals with a variety of liver disorders. However possible applications of HSA in the diagnosis of early liver damage have not bee explored. The purpose of this work was to induce liver damage in rats and detect circulating HSA in the sera. Hepatic antibodies against hepatocyte antigens were obtained immunizing rabbits with total rat hepatocytes as described elsewhere. Acute liver damage was induced with CCl4 in adult Sprague/Dawley rats. Sera from intoxicated rats and controls were studied with double immunodiffusion, immunoelectrophoresis, cross over electrophoresis, immunoelectrophoresis bidimensional and complement fixation. The results revealed the presence of circulating hepatic antigens as early as 12 hours after CCl4 administration. Complement fixation and bidimensional immunoelectrophoresis proved to be the most useful to detect early hepatic damage.
