Development of the embryonic mammalian pancreas the relationship between morphogenesis and cyto differentiation

The degree to which cytodifferentiation is obligatorily coupled to ongoing morphogenesis was analyzed in the developing mouse pancreas system. Protodifferentiated pancreatic epithelia were isolated from investing mesoderm and were cultured on plastic substrata. Within 24 h, the epithelia spread into 2-dimensional monolayers. Acinar formation failed to take place. The cells did not undergo mitosis, possibly due to the absence of mesenchymal cells or mesenchymal factor from these cultures. In spite of the absence of morphogenesis, cytodifferentiation took place on schedule. Over 5-7 days of culture, amylase specific activity rose 100-fold and zymogen granules appeared in the cells. The regulation of exocrine cytodifferentiation was independent of ongoing morphogenesis.
