Successive cumulative concentration-response curves for the contractile action of norepinephrine (NE) were constructed for isolated aortic strips prepared from untreated and reserpinized rabbits. Aortas of reserpinized rabbits were initially more sensitive to NE; i.e., lower EC50 [mean effective concentration] and greater capacity of contraction. Successive applications of NE to the aortas (i.e., sensitization of the aortas) of untreated rabbits produced a decrease in EC50 and an increase in the capacity of contraction while such a development of the response to NE was not evident with aortas of reserpinized rabbits. To make valid comparisons among agonists of NE-receptors using aortic strips, it seems necessary to subject these preparations to an identical degree of sensitization.
