Abstract
The effect of caffeine on the frequency of chromosomal aberrations induced by cis-dichlorodiammineplatinum (II) and its trans-isomer was studied. These mutagens with a delayed effect differ considerably in their effectiveness, which correlates with the fact that only the cis-isomer shows the anti-tumor activity. The protective effect of caffeine was found when caffeine (1, 0.4 and 0.08 mg/ml) and the mutagens (cis-Pt(II)- 0.01 mg/ml; trans-Pt(II) - 0.01, 0.1 and 0.2 mg/ml) were applied simultaneously during 1 h: the frequency of aberrations was 0.21-0.37 (P > 0.99) of that observed in the case of cis-Pt(II) alone and the frequency decreases in the case of the trans-isomer was not significant (0.90 < P < 0.95). Since fast interaction of caffeine and cis-Pt(II) was not found, the protective effect may be due to either the caffeine inhibiting interaction between cis-Pt(II) and DNA or the inhibition of cis-Pt(II) transport into the cell nucleus. If caffeine is applied during 1 h after the mutagens have been rinsed off, the decrease in the frequency of aberrations is not significant. If caffeine is applied during 5 h after the mutagens have been rinsed off, the frequency of aberrations strongly increases: it is 6.7-7.6 times higher than the effect of cis-Pt(II) alone and 3.9-10.2 times higher than that of trans-Pt(II). The similar response to the caffeine treatment in respect of the change in the frequency of chromosomal aberrations induced by both cis- and trans-isomers suggests that the difference in their cytogenetic effect is not due to the different capacity of cell repair system to remove DNA damages induced by these complexes.
