Effect of fluoride on pigmentation and iron distribution in the enamel of rat incisors

This study was undertaken to reveal the changes in pigmented striations and iron distribution in the enamel of rat upper incisors under various fluoride administrations. Male Wistar rats divided into 4 groups received water containing 0, 25, 50 and 100 ppm fluoride respectively for 10 weeks. Matured enamel surfaces were observed under a dissecting microscope, comparing with iron distribution analysed at identical sites by using electron microprobe. Pigmented striations appeared periodically, consisting of brown pigmented bands alternating with lighter pigmented bands on the enamel surface in every group receiving fluoride. These striations in both groups receiving 50 and 100 ppm F were thick and irregular, with widths of about 480-500 .mu.m, while those in the group receiving 25 ppm F were fine and regular. The enamel surfaces in the group receiving 100 ppm F had less pigmentation than those in the other two fluoridated groups. Line scanning analysis indicated that iron concentration was higher in the outermost enamel and decreased markedly toward the interior of the tissue. However, the peak concentration of iron moved inside, decreasing with increasing intake of fluoride. Therefore, iron distribution in the enamel related to the visible finding of the enamel surface. It was concluded that the nature of enamel formed under fluoride administration influenced the appearance of pigmented striations and the change of iron distribution.
