Effect of prostaglandin f 2 alpha and prostaglandin e 2 on the sensitivity of rabbit cortical neurons to acetyl choline and norepinephrine

Effects of microiontophoretically applied prostaglandin[PG]F2.alpha. and PGE2 on the activity pattern and sensitivity of single neurons to acetylcholine and norepinephrine were investigated in the rabbit somatosensory cortex. PGF2.alpha. generally inhibited and PGE2 increased the firing rate of the neurons. PGF2.alpha. usually changed the sensitivity of the neurons' acetylcholine, while PGE2 realized its effects upon norepinephrine-sensitive units. The influence of PGF2.alpha. on the excitatory effects of acetylcholine and the influence of PGE2 on norepinephrine inhibition were characterized as a rule by decreasing the mediator actions or by changing the initial responses sign to the opposite one. PG of group F apparently participate in cholinergic transmission while those of group E participate in adrenergic transmission. The investigated PG action is likely to be realized by selective inhibition of the adenylate or guanylate cyclase activity with subsequent decrease in synthesis of cAMP and cGMP.
