Effects and preincubation on fertilization capability and preimplantation development of preovulatory oocytes in mice

Preovulatory oocytes were obtained from the ovaries of C57BL/6J matured mice which were pretreated with PMSG and HCG respectively 58 and 10 hrs before collection of the oocytes. Morphological study of the oocytes revealed that although there exist various stages of maturation divisions of the oocytes, the proportion of oocytes with metaphase II was only less than 3%. When oocytes were inseminated with epididymal spermatozoa obtained from ICR: JCL matured mice, which were capacitated by preincubation for 1.5 hrs before use, the proportions of pronuclear ooctyes and blastocystes respectively 5 and 101 hrs after insemination were found to be low. When the oocytes were incubated with different incubation times including 2, 4 and 6 hrs, it was found that the longer the incubation time, the greater was the increase in the number of oocytes with metaphase II. Results also indicated that the proportions of pronuclear oocytes nad blastocystes encountered respectively 5 and 101 hrs after insemination apparently increase, demonstrating that appropriate preincubation of the preovulatory oocytes improved the fertilization capability and the preimplantation development of preovulatory oocytes in mice.
