Electrophoretic study of buffalo Bubalus bubalis semen


, : Electrophoretic study of buffalo Bubalus bubalis semen. Indian Journal of Experimental Biology 16(11): 1185-1187

Using discontinuous disc electrophoretic techniques, the acrosomal, sperm sonicate and seminal plasma's proteins and enzymes, i.e., acid phosphatase (ACP), acrosin, lactate dehydrogenase (LDH), malate dehydrogenase (MDH), glucose-6-phosphate dehydrogenase (G-6-PDH) and isocitric dehydrogenase (ICDH) were fractionated. Acrosomal, sperm sonicate and seminal plasma fractionated on polyacrylamide at pH 9.5 revealed 10, 12 and 11 bands, respectively. Acrosin activity was localized in the 4th, 8th and 9th of the total 11 protein fractions in the pH 4.3 system. Acrosomal ACP had 2 isozymes at the 1st and 5th positions. Seminal plasma ACP had 3 isozymes. Sperm's LDH, MDH, G-6-PDH, ICDH and ACP have 4, 2, 3, 3 and 2 isozymes, respectively. The physiological significance of these isozymes was discussed.
