The sarcotubular system [ST] of the muscle cell is a membrane system ensuring the transformation of signals, release of Ca2+ during contraction and its active uptake during the relaxation phase of the muscle. We developed a preparative electrophoretic method for separation of ST vesicles based on particle size and charge. Membrane vesicles were separated to at leat 5 morphologically and biochemically different fractions. The high content of lipids, morphological features and the extensive binding of 3H-ouabain in fraction 1 indicates that the method can be used to purify the sarcoplasmic reticulum and to separate from it a fraction enriched in plasma membrane.
