Electrophysiological and histochemical studies of cis-platinum ototoxicity were performed in guinea pigs in order to clarify its etiology and to furnish data for the prevention of ototoxicity. When the Preyer's reflex of the cis-platinum applied guinea pigs disappeared, their EP decreased to 81% of the original level but CM and N1 decreased to 15.6 and 30.1% respectively. Histochemically, a marked decrease of the nitro-BT reducibility occurred in outer hair cells. This decrease was more prominent towards the upper turns of the cochlea and towards the inner rows of outer hair cells. It is clear from these results that the early stage of cis-platinum ototoxication arises from dysfunction of the outer hair cells.
