Embryogenetic capacity of anthers from flowers formed in vitro on thin cell layers and of anthers excised from the mother plant of nicotiana tabacum cultivar samsun and nicotiana plumbaginifolia

In vivo anthers (from entire plants) and in vitro anthers (induced from thin layers of epidermal and subepidermal cells) of both N. plumbaginifolia and N. tabacum were compared for the embryogenetic capacity of their pollen grains. Both types of in vivo and in vitro pollen grains were capable of forming haploid embryos when cultured on basal medium of Murashige and Skoog additioned or not with IAA (10-6 M) and kinetin (10-6 M). N. plumbaginifolia in vitro anthers yield a higher percentage of haploids than in vivo (20% for in vitro vs. 3% for in vivo). This higher capacity of embryo formation shown by in vitro pollen grains seemed to be correlated with the higher frequencies of chromosome combinations in pollen anther cells.
