Endorphins in several regions of rat brain large differences between radio immunoassay and radio receptor assay

Various endorphins (i.e., peptide fragments from .beta.-lipotropin) with affinity for the opiate receptors were characterized. Two pentapeptides, methionine-enkephalin (Met-ENK) and leucine-enkephalin (Leu-ENK) were found in brain, and larger fragments, the .alpha.- and .beta.-endorphins (61-76 and 61-91 residues, respectively) in pituitary. Marked regional differences in total endorphins (RRA [radioreceptor assay]) and Met- and Leu-ENK (RIA [radioimmunoassay]) were observed, with similar ranking, for the levels of the 3 groups of compounds when using HCl extraction: highest in striatum and hypothalamus and lowest in hippocampus and cerebellum. Total ENK levels (RIA) in each region represented only 3-20% of the total endorphins (RRA). Because, in contrast with the opiate receptors which have similar affinities for all classes of endorphins, these antisera against ENK did not recognize the larger classes like .alpha.- or .beta.-endorphins; this difference was interpreted as evidence for the presence of significant amounts of the latter compounds in brain regions and not only in pituitary, as previously thought.
