Evaluation of microfilaremia loa loa filariasis using the calibrated thick drop method influence of some technical parameters

Microfilariae were examined from venous blood samples from 6 patients with L. loa microfilariae. The influence of several technical parameters on the evaluation of microfilaremia were studied using the calibrated thick drop method: the size of the smear, the duration of drying and Hb removal. This was done by staining with a 1 in 20 dilution of Giemsa and comparing the microfilarial densities with the results of direct examination. The best protocol is as follows: clean and ungreased slides, smear of 3 cm2, prolonged drying (24 h minimum), and direct staining without initial hemoglobin removal. In these conditions there is no loss of microfilariae, the values being the same as those obtained on fresh smears without staining.
