Evaluation of spermatozoan mitochondrial activity with reference to motility and livability among holstein friesian bulls and murrah buffaloes
The spermatozoan mitochondrial activity in relation to initial motility was studied. Cytochemical method (NBT reductase test) was employed to assess the dehydrogenase activity taking place within the sperm mitochondria. Phase contrast optics were used for scoring both the characteristics referred to. Nine semen samples from 6 Holstein-Friesian bulls and 12 semen samples from 6 Murrah buffaloes were considered. The extent of deposition of the reduced dye within the mitochondrial sheath was used to assess the dehydrogenase activity and thereby the livability. The stain deposition was quantitated with the aid of phase contrast microscopic observations.
