The existence of a PIF [prolactin inhibiting factor] separate from DA [dopamine] was confirmed by assaying in vitro the prolactin-inhibiting activity of [rat] hypothalamic extracts from which catecholamines were eliminated by adsorption on Al2O3, and by testing the prolactin-inhibiting activity of such Al2O3-treated or untreated extracts and of DA in presence of DA receptor inhibitor. The maintenance of prolactin-inhibiting activity in the synaptosomal after adsorption of catecholamines, and the failure of haloperidol to significantly antagonize the effects of PIF in this fraction, strongly support the hypothesis that another hypothalamic factor besides DA can inhibit the release of prolactin from the pituitary.
