An experimental model allowing evaluation of antianginal drugs in myocardial areas either normal or made ischemic by temporary coronary occlusion was developed in the dog. This model associates the mapping of regional myocardial blood flow (endo and epicardial) by labeled microspheres and the measurement of epicardial ST-segment. A 12 min occlusion of a diagonal branch of the left anterior descending coronary artery induces an heterogenous distribution of blood flow at the ischaemic areas level, the reduction observed being most marked in the endocardium. Significant ST-segment elevation was only observed in areas where regional blood flow (epi + endo) was reduced by at least 50%. All hemodynamic and electric changes induced by the 12 min coronary occlusion have disappeared 45 min later. A 2nd occlusion, similar to the 1st, and realized after recovery of all parameters, induces almost identical hemodynamic and electric modifications showing the reproducibility of the model.
