One biochemical change observed in inflammation and tumor state is the increase of serum haptoglobin level. Our serial studies have demonstrated that many inflammatory agents induce hyperhaptoglobinemia. Thus, to elucidate the mechanism of hyperhaptoglobinemia on the molecular level, transcriptional level of rabbit haptoglobin gene was determined by hybridization of the rabbit poly (A)+ RNA isolated from the inflammatory agent-treated liver using radiolabeled probe, 643 bp of .beta.-region in human haptoglobin .alpha.2.beta. cDNA. Transcriptional level was increased maximally 12 hours after injection. Turpentine and endotoxin were more potent than carrageenan and prostaglandin E1 in elevating the transcriptional level of rabbit haptoglobin gene. Based on these results, it can be said that the above described three agents except prostaglandin E1 may induce hyperhaptoglobinemia by enhancing the transcription in addition to the massive release of haptoglobin stored in the liver. Prostaglandin E1 may induce the increase of haptoglobin biosynthesis by stimulation of translation reaction.
