Fluorescence characteristics of sodium fluorescein in plasma and whole blood

In vitro measurements were made of the excitation and emission spectra of thin layers of plasma and whole blood [human] containing sodium fluorescein. Measurements were performed using frontal illumination of the sample: that is, the emitted fluorescence was measured from the side of the sample which was illuminated by the excitation light beam, thereby simulating the conditions of fundus angiography. The influence of sodium fluorescein concentration, layer thickness, hematocrit and O2 saturation of the blood on the fluorescence characteristics was investigated. The experimental results were compared with a theoretical analysis of the effect of absorption by blood on the fluorescence of sodium fluorescein diluted in blood.
