Varying doses of 1.alpha.-hydroxycholecalciferol (1.alpha.-HCC) (50, 150, 250 and 350 .mu.g) in propylene glycol were injected i.m. into 30 dry adult Israeli Friesian cows. Four animals recieved a 2nd dose (250 or 350 .mu.g) 48 h later or ten (350 .mu.g) 72 h after the 1st dose. Plasma Ca rose after 24 h at all dose levels except 50 .mu.g. A dose-dependent peak in plasma Ca was reached after 3-4 days, followed by a return to baseline 5 days (150 .mu.g) and 8 days (250 and 350 .mu.g) postinjection, respectively. Repeating the injection 48 or 72 h later increased the time span by 3 or 4 days, respectively. The effect on plasma inorganic phosphate was double that on plasma Ca. Plasma Mg declined slightly 3 days postinjection. High Ca feeding in conjunction with 1 or 2 injections of 350 .mu.g 1.alpha.-HCC did not modify the response of plasma Ca. An injection of 350 .mu.g of 1.alpha.-HCC was given once to 40 parturient-prone cows of the same breed, and twice, at 72 h intervals, to 37 such cows. Six of these animals received 5 mg of flumethasone together with the 2nd injection, and 13 received it 48 h later, to induce parturition, which occurred 24-48 h postinjection. None of the cows injected earlier than 24 h prepartum developed parturient paresis in comparison with 22 of 60 control animals which developed the condition. There were no local or systemic clinically detectable signs of toxicity in any of the injected animals. Apparently 1.alpha.-HCC is a valuable agent in the prevention of bovine parturient paresis.
