Gas liquid chromatographic analysis of sugar and sterol fractions for characterizing uredospores of yellow rust puccinia striiformis brown rust puccinia hordei and conidia of powdery mildew erysiphe graminis

Analyses of special sugar and sterol fractions from uredospores of Puccinia striiformis WEST. and Puccinia hordei OTTH. as well as from conidia of Erysiphe graminis DC. by gas-liquid chromatography are presented. After hydrolysis of uredospores nearly 40% by weight of the spores could be identified as monosaccharides. The fraction of water soluble sugars is composed mainly by the sugar alcohols arabitol and mannitol. Mannose and a few amounts of glucose result from the analysis of the polymer fraction. Regarding the conidia there are more water soluble glucose and fructose additional to arabitol and mannitol than in the rust spores. Moreover the conidia contain trehalose, which could not be detected in dormant but in germinated uredospores. Using the ratio of arabitol to mannitol one can distinguish the spores of yellow rust from that of brown rust and powdery mildew. Qualitative and quantitative differences were shown to exist between the sterol fractions from uredospores of Puccinia striiformis WEST. and Puccinia hordei OTTH. It was impossible to draw a distinction between races and formae speciales of one and the same fungal species by means of the sterol and sugar patterns.
