Histochemical studies on the activity of atpase and auto radiographic study on the uptake of radio carbon atp in ascaris lumbricoides suis

Histochemical studies were carried out in order to identify the distribution of ATPase in A. lumbricoides suis. The worms were cultured in the incubator at 37.degree. C in Ringer-Dale solution for 3 days, then immersed in cold acetone solution and kept in a freezer at -70.degree. C. They were sectioned in a cryostat at 8.0 .mu. thickness and stained by the Padykula-Herman method. Microscopically the ATPase was located in subcuticular syncytial cells, striated epithelium of the intestine and reticular cells. Uptake of 14C-ATP was studied by microautoradiography. The worms were kept in Tyrode's solution containing 14C-ATP for 1 h at 37.degree. C, sectioned in a cryostat and examined after the Fitzgerald stripping microautoradiography. The label was concentrated in the epithelial cells of the intestine and the subcuticular tissues.
