Immunofluorescence localization of plum pox virus and prunus necrotic ringspot virus in tissue sections of prunus cerasus and prunus juxa

The distributions of plum pox virus and prunus necrotic ringspot virus (PNRV) in Prunus juxa and P. cerasus was determiend with the aid of immunofluorescence labelling. Only few shoot tips of field-grown plants exhibited a small virus-free region of the apical meristem. Most of the shoot tips were completely virus-infected. Results of in situ immunolocalization performed on tissue both from cultures and from field-grown plants are in agreement with ELISA. Results of both methods indicate that the complete elimination of PNRV from P. cerasus cannot be achieved solely by culturing tissue from shoot tips.
