In vitro culture of nicotiana tabacum anthers part 2 cyto genetic analysis of callus tissue derived from anthers by prolonged sub culture

The primary callus of 'Samsun' tobacco is characterized by dominance of haploid, diploid and aneuploid cells. The subculture of tissue culture strains in 1 yr showed shifts in frequencies of cells with different ploidy and the formation of haploid, diploid and tetraploid strains at the end of culturing. Multinuclear cells and extrusion of chromosome material were seen. The frequency of chromosome aberrations in primary callus and in cells of haploid and diploid strains was 6-16%. The tetraploid strain of callus tissue had a higher level of chromosome aberrations.sbd.40%. A possible mechanism for the formation of strains of different ploidy is discussed.
