Industrial styrene poisoning 10. determination of styrene in biological materials by gas chromatography

For monitoring solvent exposure, it is useful to determine metabolites of solvents in urine and solvents themselves in blood and tissues. Aliquots of 0.5 g of tissues with 5 ml of n-hexane added were homogenized by a high-speed homogenizer (Polytoron) for 10-30 s and the filtrates containing extracts were analyzed for styrene by gas chromatography. Linearity was ascertained from the calibration curve obtained by adding the known quantities of styrene (4, 10, 20, 40, 100 ppm) to blood, liver and adipose tissues of rats. Rates of recoveries of styrene were 92-101%. Reproducibility was examined by repeated determinations of styrene in blood, liver and adipose tissues of rats exposed to 500 ppm styrene for 4 h. Coefficients of variation were 2.8-14.0%. There was an approximately linear relationship between styrene concentration (0-1000 ppm) of exposed air and blood of exposed rats. This simple and rapid method was applicable to determination of solvents other than styrene in organs and tissues by combining suitable solvents for extraction and packing of gas chromatography.
