Isolation of dark tobacco nicotiana tabacum breeding lines differing in alkaloid concentration via anther derived haploids

Two sources of low alkaloid genes were used to establish low and intermediate alkaloid genotypes in the normally high alkaloid dark tobacco (Nicotiana tabacum L.) background. Haploid plants were generated via anther culture from five advanced breeding lines of dark tobacco segregating at loci controlling alkaloid production. Haploid plants were screened for alkaloid concentration and diploids of selected haploids were obtained using midvein explants. Progeny from one hundred and seventy-two doubled haploids were grown in replicated field plots and analysed at harvest for total nitrogen, nitrate nitrogen, and total alkaloid concentration. Total nitrogen and nitrate-N showed little variation among the breeding lines or among alkaloid classes within a breeding line. Dark tobacco doubled haploid lines ranging from 0.56 to 5.00 per cent total alkaloid concentration on a dry weight basis were identified. These results indicate that: 1. the control of alkaloid concentration in dark tobacco is similar to that in burley and flue-cured tobacco, and 2. tissue culture techniques can be used in conjunction with conventional breeding programs to select for alkaloid characteristics derived from both quantitative and qualitative genetic sources.
